Course XI. Physics, chemistry and biology of bioluminescence



DATOS SOBRE LA ACTIVIDAD

OBJETIVOS:

The aim of this course is focused on exploring basic and applied aspects of
bioluminescence.

Priority objectives. After successfully completing the course participants:

- shall be able to understand basic mechanisms of bioluminescence of various
luminous organisms;

- shall have a good knowledge of the main principles and methods of bioluminescent
analysis and their application in biology, ecology, medicine etc.

Monday 26 September

8h 30 - 9h Welcome to students.

9h – 11h Lecture 1. Overview of bioluminescence: bioluminescence in nature, luciferases and luciferins, photoproteins, biochemistry and biophysics of bioluminescence, spectra in bioluminescence Speaker: Prof. Valentina Kratsyuk (SFU, Russia)

11h – 11h 30 Coffee Break.

11h 30 – 13h 30 Lecture 1.1 Determination of the main kinetics parameters of NAD(P)H:FMN-oxidoreductase + luciferase bioluminescent system; Speaker: Oleg Sutormin (SFU, Russia)

Tuesday 27 September

9h – 11h Lecture 2. Description (Enzyme and substrate/co-substrate nature, Enzyme commission number) of different luciferases (Firefly/bacterial/Cypridina/Fungi/ Renilla/ Gonyaulax/ Gaussia/ Metridia/ Vargula/ Cypridinaluciferase), and respective luciferins (structure). Photoproteins. Speaker: Prof. Valentina Kratsyuk (SFU, Russia)

11h 30 – 13h Lecture 2.1 Determination of the kinetics features of enzyme reactions (Km, Vmax) using NAD(P)H:FMN-oxidoreductase + luciferase bioluminescent system. Speaker: Oleg Sutormin (SFU, Russia)

Wednesday 28 September

9h – 11h Lecture 3. Cloning of different bacterial lux gene cassette and their various applications in healthcare, basic research and bio-diagnostics. Speaker: Prof. Valentina Kratsyuk (SFU, Russia)

11h 30 – 13h Lecture 3.1 Determination of pH and temperature optimums of NAD(P)H:FMN-oxidoreductase + luciferase bioluminescent system. Speaker: Oleg Sutormin (SFU, Russia)

Thursday 29 September

9h – 11h Lecture 4. Terrestrial bioluminescence: firefly and insects, bacteria, fungi, mollusks, arthropods and earthworms. Speaker: Prof. Valentina Kratsyuk (SFU, Russia)

11h – 11h 30 Coffee Break.

11h 30 – 13h 30 Lecture 4.1 Analysis of activity of NADH-dependent dehydrogenases (lactate dehydrogenase, glucose-6-phosphate dehydrogenase, alcohol dehydrogenase). Speaker: Oleg Sutormin (SFU, Russia)

Friday 30 September

9h – 11h Lecture 5. Bioluminescent analysis. Speaker: Prof. Valentina Kratsyuk (SFU, Russia)
